The MicroVue CICP assay measures type I C-terminal collagen propeptide (CICP) in serum. CICP may be useful as a biochemical indicator of collagen production and growth.

The Marker
Collagen, the triple-helical molecule which forms the fibrous framework of all connective tissues, is synthesized as procollagen, a larger precursor molecule. Procollagen consists of mature collagen with extension peptides at both the amino and carboxy termini. These extension peptides, or propeptides, are cleaved from the collagen molecule by specific proteases prior to incorporation of collagen into a growing collagen fibril. The release of these peptides into the circulation provides a stoichiometric representation of the production of collagen.

Assay Steps |
- Add 100 µL Standards, Controls and 1:12 diluted samples
- Incubate 120 ± 5 minutes at 18-25°C
- Wash 3 times with 1X Wash Buffer
- Add 100 µL Rabbit anti-CICP
- Incubate 45-50 minutes at 18-25°C
- Wash 3 times with 1X Wash Buffer
- Add 100 µL Enzyme Conjugate
- Incubate 45-50 minutes at 18-25°C
- Wash 3 times with 1X Wash Buffer
- Add 100 µL Working Substrate Solution
- Incubate 30-35 minutes at 18-25°C
- Add 50 µL Stop Solution and read Optical Density at 405 nm
